| Inhibition Assay of Dengue Virus |
Description: | Vero E6 cells (BCRC #60476, Bioresource Collection and Research Center, Hsinchu, Taiwan) were treated with one of Compounds 1-9 at a predetermined concentration for 1 hour at 37° C. The cells were infected with Dengue virus type 1 (DV1, ATCC VR-1856, American Type Culture Collection, Manassas, VA, USA) at MOI=0.002 or Dengue virus type 2 (DV2, ATCC VR-1584, American Type Culture Collection, Manassas, VA, USA) at MOI=0.02, both for 5 days at 33° C. They were then fixed with 10% formalin and permeabilized with methanol. Subsequently, the cells were stained with anti-flavivirus E antibody (ATCC HB-112, American Type Culture Collection, Manassas, VA, USA) and anti-mouse IgG-FITC (MP Biomedicals, Irvine, CA, USA). The nuclei of the cells were counter stained with DAPI (Thermo Fisher Scientific, MA, USA). The E protein expression was measured by using a high-content image analysis system (Molecular Devices). The cell viability was determined by MTS. EC50 and CC50 were calculated by Prism software. |
