| Description: | The test compounds were dissolved in assay buffer (50 mM Hepes, 150 mM NaCl, 5 mM CaCl2, pH 7.4) at a final concentration of 10 μM and 1 μM. Coagulation factor VIIa and tissue factor were mixed at an equimolar concentration. After incubation at 37° C. for 15 minutes, the test compound was further added. Incubation was conducted at room temperature for 10 minutes. Then the substrate (Boc-VPR-AMC) was added to initiate the reaction. For reading fluorescence signal value, enzyme kinetic mode was used. The wavelength of excitation light was 380 nm, and the wavelength of emission light was 460 nm. |
