| KRAS G12D/SOS1 Binding Assay |
Description: | (1) Compound was prepared with DMSO, and diluted by 3-fold gradient with DMSO.(2) 0.1 μl of serial dilutions of the compound was added to a 384-well plate.(3) 5 μl of Tag2-KRASG12D>P at a specific concentration was added to the 384-well plate, and the plate was centrifuged at 1000 rpm for 1 min.(4) Then 5 μl of Tag1-SOS1 at a specific concentration was added to the 384-well plate, and the plate was centrifuged at 1000 rpm for 1 min.(5) The plate was incubated at 25° C. for 15 min.(6) 10 μl of the mixture of anti-Tag1-Tb3+ and anti-Tag2-XL665 was added to the 384-well plate.(7) The plate was centrifuged at 1000 rpm for 1 min and incubated at 4° C. for 3 h.(8) The 665/615 nm ratio was read by a microplate reader.(9) Data analysis: The log value of the compound concentration was taken as the abscissa, and the 665/615 nm ratio was used as the ordinate, the data were analyzed by GraphPad Prism 8.0 software. |
