| Cathepsin Ezyme Assay |
Description: | Cathepsin B and H were first activated in the presence of thiol activators at pH 6.0 and pH 7.0, respectively. Then, 15 µL of the enzyme solution was mixed with 940 µL of 0.1 M sodium phosphate buffer containing 1 mM EDTA separately, for 10 min at 37 °C. Then, 20 µL of stock solution (5 mM) of different compounds were added separately to the activated enzyme assay mixtures to effect final drug concentrations as 1 × 10^-4 M in 1 mL assay (4.5% DMSO). After 30 min, 25 µL of 100 mM substance stock solution was added to start the reaction. |
