| Description: | In vitro inhibition assay of human PFT was done by measuring the incorporation of [3H] FPP (Amersham Biosciences, Piscataway, NJ) into wild type H-Ras (PFT). The 60,000g post-microsomal supernatant from Daudi cells was incubated in the presence of increasing concentration of compound, H-Ras substrate, and [3H] FPP. Samples were TCA-precipitated and then filtered onto glass fiber filters; unbound [3H] FPP was washed through the filters. Samples were counted on a scintillation counter and activity compared to vehicle controls to obtain IC50 values. |
