| Description: | The direct CYP1A1 inhibitory activity of test compounds was also assessed using the Promega P450-Glo assay system. Seven concentrations of test compound were added to a ½ area white 96 well plate. Cypex CYP6A. bactosomes ([final] 0.5 pmol) and CYPA substrate Luciferin-CEE ([final] 30 μM) were prepared in 0.1M potassium phosphate buffer and incubated with test compounds at 37° C. for 5 minutes. 0.2m NADPH was then added to the plates and incubated at 37° C., for 10 minutes. The reaction was stopped by adding luciferin detection reagent and luminescence was read after 20 minutes. |
