Description: | Briefly, the methods were: On the day before the assay 20,000 cells per well of CHEM1-OGR1 cells were plated in 10% DMEM containing 10% FBS and 1% Pen/Strep into a 96 well plate. After 24 hours media was removed and cells were stained with 80 uL Fluo-8 calcium indicator dye according to manufacturer's protocol. After staining cells were treated with 20 uL of 4× concentration of test compounds. Using kinetic imaging in the Lionheart HCS fluorescence intensity was continuously measure at 10 frames per second, for 5 seconds before 100 μL of acidified media was added to the well resulting in a pH of 6.4. A ratio of fluorescence intensities prior to addition and at peak were calculated and averaged. The resulting data were platted and using a 4 parameter logistic regression EC50 for each test compound was determined. |
