Assay Name:  In Vitro Binding Test of Compound and PSMA Protein
Description:  A Biacore 8K (Cytiva) instrument was used to determine the binding affinity of disclosed compounds to the PSMA protein (Sinobiological). The PSMA protein was captured on SA chips. Before immobilizing the disclosed compounds (flow path 1 and 2, flow rate 10 μL/min), the PSMA protein was immobilized on flow path 2 by using flow buffer (10 μg/ml, flow rate 5 μL/min, injection time 600 s), and 1M NaCl was injected three times consecutively into 50 mM NaOH to condition the sensor surface. After each disclosed compounds injection, isopropanol in 1M NaCl and 50 mM NaOH was used for additional washing (flow path 1, 2, flow rate 10 μL/min, injection time 60 s). All compounds were dissolved in 100% DMSO and diluted to 10 mM, and then diluted in test buffer (PBS, pH 7.4, 1 mM TCEP (tris-(2-hydroxyethyl)phosphine), 0.05% P20, 2% dimethyl sulfoxide) at an appropriate highest concentration.
