| Description: | Assay: Compounds are dissolved in DMSO and diluted to 10 mM concentration in DMSO immediately before IC50 analysis. To determine inhibition of the enzyme, 10 μL of DMSO was added to the first cuvette, and 10 μL of test compound stock solution was added to the second cuvette. Enzyme was then added to the cuvettes. The IC50 experiment was performed for the following test compound concentrations: 20, 10, 3, 1, 0.3, 0.1, 0.03, and 0.01 μM. If the test compound was more potent than 0.3 μM, the concentrations used in the IC50 determination were shifted to lower values such as: 3, 1, 0.3, 0.1, 0.03, 0.01, 0.003, and 0.001 μM. |
