| Inhibition of Compounds on Viral Replication |
Description: | Determination of the inhibitory activity of compounds on the replication of 2019 novel coronavirus (SARS-CoV-2): anti-novel coronavirus activity test methods of NHC and GS-441524 are as reported in the literature. To Vero cells infected with the 2019 novel coronavirus, different concentrations of test compounds were added. After 48 hours of incubation, the virus copy number in the cell supernatant was quantified by quantitative real-time RT-PCR (qRT-PCR) to evaluate the inhibitory activity of the compound on the virus. Determination of the inhibitory activity of compounds on the replication of respiratory syncytial virus (RSV), human coronavirus OC43, influenza A virus, and Zika virus by Cytopathogenic effect (CPE): the experimental cells were inoculated at a certain cell density to 96 well cell culture plates, and cultured overnight in a 5% CO2, 37° C. incubator. The compounds and viruses were added the next day. Depending on the tested virus, the cells were cultured in an incubator for 3-7 days under 5% CO2, 33° C. or 37° C., until 80-95% of the cells in the virus-infected control wells without compound had pathological changes. Cell viability in each well was then detected with CellTiter-Glo or CCK-8. If the cell viability of the compound-containing well is higher than that of the virus-infected control well, that is, the CPE is weakened, it indicates that the compound has an inhibitory effect on the tested virus. |
