| Description: | For inhibitor testing, the sample composition was the same as described above, except for the addition of the putative inhibitory compound. This resulted in presence of 1% or 2% (v/v) DMSO in reaction mixture. Inhibitory constants were determined using different concentration of H-Gln-AMC varying from Km-2 Km. Final concentration of bacterial QC was in a range between 25 nM-50 nM. The inhibitory constant was evaluated by fitting the set of progress curves to the general equation for competitive inhibition using GraFit software (Version 7, Erithacus software Ltd., Horley, UK). |
